NO Binding Dynamics to Hemoglobin

NO binding rates to hemoglobin is important in sickle cell research. To measure these rates, we used a flow-flash setup to mix carboxy-hemoglobin with NO buffers of various concentrations. The CO was then photolyzed off the hemoglobin with an intense laser pulse (355 nm, 7 ns, ~15 mJ/cm2) and the NO binding was observed through absorption spectroscopy with the streak camera.
